Complex retroviruses, retroelements, HIV p24 and their hosts

Epigenetic therapy of myelodysplastic syndromes connects to cellular differentiation independently of endogenous retroelement derepression.

myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) characterized by abnormal epigenetic repression and differentiation of bone marrow haematopoietic stem cells (HSCs). Drugs that reverse the epigenetic repression, such as 5-azacytidine (5-AZA), induces hematologic improvement in half of the patients treated.

Although the underlying mechanisms of therapeutic efficacy is not yet clear, the induction of endogenous retroelements (eres) has hypothesised.Using RNA sequencing (RNA-seq), we compared the transcriptional Eres in bone marrow HSCs of a new cohort of MDS and chronic myelomonocytic leukemia (CMML) patients before and after 5-AZA treatment with HSCs from healthy donors and patients with AML.

We further examined the ERE transcription using a comprehensive explanation ERE partially-overlapping transcripts expressed in HSCs, produced here by de novo assembly and supported by transcripts of full-length RNA-seq.Consistent with previous reports, we found that treatment with 5- AZA increased representation of ERE-derived RNA-seq reads in the transcriptome. However, the increase is comparable between treatment response and failure.

Extended view HSC transcription diversity offered by de novo assembly transcripts against Eres 5-AZA-responsive as a determinant of treatment outcome. In contrast, pre-treatment revealed expression and alternative splicing gene transcripts regulated as a predictor of response to the development of MDS and CMML patients to 5-AZA treatment.Our studies identify transcriptional signature-coding genes regulated development of non-protein-coding and not eres, as correlated good response from MDS and CMML patients for the treatment of 5-AZA and offer new candidates for further evaluation.
